以钌(Ⅱ)多吡啶配合物[Ru(bipy)2DAFND](Cl O4)2(Ru-DAFND,bipy=2,2'-联吡啶,DAFND=4,5-二氮杂芴-9-对硝基苯腙)为指示剂,采用锁相放大技术构建了基于荧光猝灭原理的光纤DNA传感器,研究了传感器的性能。以p H 7.1的Tris缓冲溶液为介质,在2.6×10-8~5.4×10-6mol/L范围,ct DNA浓度与传感器光学敏感膜的相对滞后相移(Δφ)有较好的线性关系,检出限为8.4×10-9mol/L,响应时间为70 s。通过研究溶液p H值和干扰物对传感器性能的影响,表明该传感系统具有较好的重复性和稳定性。 相似文献

Reverse Watson–Crick DNA with parallel‐strand orientation (ps DNA) has been constructed. Pyrrolo‐dC (PyrdC) nucleosides with phenyl and pyridinyl residues linked to the 6 position of the pyrrolo[2,3‐d]pyrimidine base have been incorporated in 12‐ and 25‐mer oligonucleotide duplexes and utilized as silver‐ion binding sites. Thermal‐stability studies on the parallel DNA strands demonstrated extremely strong silver‐ion binding and strongly enhanced duplex stability. Stoichiometric UV and fluorescence titration experiments verified that a single 2pyPyrdC–2pyPyrdC pair captures two silver ions in ps DNA. A structure for the PyrdC silver‐ion base pair that aligns 7‐deazapurine bases head‐to‐tail instead of head‐to‐head, as suggested for canonical DNA, is proposed. The silver DNA double helix represents the first example of a ps DNA structure built up of bidentate and tridentate reverse Watson–Crick base pairs stabilized by a dinuclear silver‐mediated PyrdC pair. 相似文献

The gelation behavior of lithocholate (LC?) with different metal ions in water was investigated. The microstructures of hydrogels were determined to be three‐dimensional (3D) networks of fibrous aggregates. The formation of fibrils was speculated to be mainly driven by the coordination between carboxylate of LC? and metal ions, accompanied by the assistance of noncovalent interactions such as electrostatic and hydrophobic interactions. The hydrogels, which can maintain the mechanical strength at higher temperature, exhibit thermal stability. Their gelation capability was enhanced with the increase in acidity. The hydrogels of LC? and Cu2+ mixtures served as the precursors for producing network nanostructures of CuS nanoparticles. These new CuS networks exhibit high fluorescence quenching ability and can act as an effective fluorescent sensing platform for ssDNA detection. 相似文献

By combining ion‐mobility mass spectrometry experiments with sub‐millisecond classical and ab initio molecular dynamics we fully characterized, for the first time, the dynamic ensemble of a model nucleic acid in the gas phase under electrospray ionization conditions. The studied oligonucleotide unfolds upon vaporization, loses memory of the solution structure, and explores true gas‐phase conformational space. Contrary to our original expectations, the oligonucleotide shows very rich dynamics in three different timescales (multi‐picosecond, nanosecond, and sub‐millisecond). The shorter timescale dynamics has a quantum mechanical nature and leads to changes in the covalent structure, whereas the other two are of classical origin. Overall, this study suggests that a re‐evaluation on our view of the physics of nucleic acids upon vaporization is needed. 相似文献

Wedelolactone (WLA) is a polyphenolic coumestan derivative found in extracts of plants used in traditional medicine. Due to its cytostatic activity, WLA is one of natural compounds tested as potential anticancer drugs. In this work we for the first time studied electrochemical properties of WLA using cyclic (CV) and square‐wave (SWV) voltammetry at the basal‐plane pyrolytic graphite electrode. A reversible pair of peaks, corresponding to catechol/o‐benzoquinone redox system, was observed using CV around 0.275 V vs. Ag|AgCl|3 M KCl reference electrode. Measurements of SWV signal of WLA in the presence of single‐ or double‐stranded DNA suggested a weak interaction without evident preference for double‐stranded DNA. An indirect assay, employing electroactive DNA intercalator doxorubicin as competitor, confirmed absence of intercalative DNA binding of WLA. 相似文献

Novel electrochemical DNA‐sensor based on glassy carbon electrode (GCE) modified with Ag nanoparticles, Neutral red covalently attached to its surface and native DNA adsorbed on modifier coating was developed for the estimation of DNA damage on example of model system based on Fenton reagent. As was shown, the oxidation process resulted in synchronous increase of electron transfer resistance and capacitance measured by electrochemical impedance spectroscopy (EIS). The contribution of each sensor component on the signal was specified and sensitivity estimated against similar surface coatings. The shift of EIS parameters was found to be higher than that of similar biosensors reported. The DNA sensor was tested on the estimation of antioxidant capacity of green tea infusions again the results of coulometric titration with electrogenerated bromine. 相似文献

Despite significant progress in development of bioanalytical devices cost, complexity, access to reagents and lack of infrastructure have prevented use of these technologies in resource‐limited regions. To provide a sustainable tool in the global effort to combat infectious diseases the diagnostic device must be low cost, simple to operate and read, robust, and have sensitivity and specificity comparable to laboratory analysis. Electrochemical monitoring of DNA hybridization related to p53 gene sequence was investigated using genomagnetic assay combined with single walled carbon nanotube (SWCNT) modified pencil graphite electrodes (PGEs). The hybridization was performed either at magnetic beads (MB) surface or in solution. The enhanced guanine signal was obtained using SWCNT‐PGEs compared to one obtained by unmodified PGEs. The selectivity of genomagnetic assay was tested under optimum conditions. The DLs were calculated as 0.88 µM and 0.11 µM for hybridization performed at MB surface and solution, respectively. This selective, practical and cost effective genomagnetic assay combined with SWCNT‐PGEs is reported herein for the first time. 相似文献
